The winners of this game will face either Juventus or Salernitana, not finding out which until that game on January 4.
Walter Mazzarri got the Partenopei back on track with victories against Cagliari and SC Braga, setting up a Champions League Round of 16 showdown with Barcelona in February.
Victor Osimhen and Kvicha Kvaratskhelia are among those rested, while Eljif Elmas, Piotr Zielinski, Andre Frank Zambo Anguissa and Mathias Olivera are not fully fit.
It is an opportunity for Simeone, Raspadori and summer signing Lindstrom to prove themselves.
Frosinone are comfortably mid-table in Serie A, but have never beaten Napoli, managing two draws in 10 competitive meetings.
This is the first time the Canarini have ever made it to the Coppa Italia Round of 16.
Luca Mazzitelli, Reinier, Riccardo Marchizza and Sergio Kalaj are injured, but Enzo Barrenechea and Walid Cheddira bring quality to the proceedings.
